1. Working principle

This instrument is based on Soxhlet extraction and weight measurement as the basic principles to determine fat content; that is, in the presence of a solvent, the fat is dissolved to extract the fat with the solvent, and the fat is separated from the solvent by extraction and dried 、 Weigh and calculate fat content.

3. Preparation before operation

3.1 Sample preparation: (All samples are subject to clean and dry conditions. Try to be as uniform as possible when sampling.)

3.1.1 Cereals: grains and beans (except peanuts), take out 30 to 50g of net dry samples to remove impurities, grind them through a 1.0mm diameter round hole sieve, and put them in jars for use.

3.1.2 Small grains of oil: such as sesame, rapeseed, etc., take out about 20g of the net dry sample with impurities removed, and put it in a wide-mouth bottle for use.

3.1.3 Large-grain oil: For example, peanut fruit, castor seeds, sunflower seeds, etc., take a net dry sample of 30 ~ 50g, peel the shells one by one, weigh the kernel shell separately, calculate the percentage of kernel weight, and then slice into the jar spare.

3.2 Wash the extraction bottle with distilled water until no impurities are left in the cylinder, place it in a drying oven and bake it for about 1 hour (the temperature is 105 ° C), take out the number, weigh it, and move it into the desiccator for cooling.

3.3 Put the cylindrical filter paper barrel into the filter paper barrel holder. (Note that the upper mouth of the filter paper must not be higher than the inner slot of the filter paper drum holder)

3.4 Check whether the power supply is intact and the water source is connected firmly.

3.5 Turn on the power switch, set the temperature of the temperature controller and the temperature adjustment knob, and preheat for 15 minutes.

4. Operation

4.1 Sample:

1. Low-fat samples such as grain and feed can be weighed directly to the nearest 0.0002 grams. First make up a layer of absorbent cotton in the filter paper bucket, then transfer the sample into the cylinder, and then cover the sample with absorbent cotton.

2. Weigh the samples of oil and other high fats and oils in a spare sample, weigh the sample, pour it into the oven, put it in a drying oven and bake at 105 ℃ for 30 minutes, then pour it into a mortar and grind it while hot. Add appropriate amount of defatted fine sand (about 2 grams) and grind it with the sample, grind the sample to an oily state (note: the sample must be oily, not sticky, and have a certain degree of looseness), Transfer cleanly into a filter paper tube plugged with absorbent cotton at the bottom, dip a little ether with cotton wool, wipe off the sample and fat on the mortar, (preferably use a small amount of ether to clean the mortar) and merge it into the filter paper tube, then Put the absorbent cotton into the upper part to press the sample and put it into the extractor.

4.2 Inject about 340ml of anhydrous ether into the extraction bottle, first put the extraction bottle on the extraction device, and move it to the water bath, adjust the position to keep the extraction bottle in good contact, and make the cock handle vertical.

4.3 Turn on the power and adjust the heating temperature as required. The display will show the heating temperature directly.

4.4 When recovering the solvent, just make the cock handle horizontal.

4.5 After recovering the ether, lift the condenser tube and remove the extraction bottle from the water bath. Place it in a constant temperature oven, bake off the water, then move it into a drying cylinder to cool it, weigh it, and calculate the oil content.

4.6 After use, turn off the power, scrub it clean, and keep it tidy.

5. Matters needing attention (common faults):

5.1. Use two sets of water sources to cool in and out to prevent the volatilization of ether;

5.3. The heating method is water bath heating. Distilled water must be injected before heating to prevent the electric heating tube from burning out.
