The summer vacation is now halfway through, and yesterday afternoon, my company's manager reviewed and analyzed several commonly asked ELISA kits that our customers often inquired about. I'm sharing this information here for your reference. 1. When performing tissue ELISA, what kind of liquid is used for tissue grinding? Shanghai Hengyuan Technical Analysis: If you're aiming to detect components within the tissue, it's recommended to use a buffer with a pH around 7 or lower, and with low ionic strength, to ensure minimal interference with the results. If you're using ground tissue for coating, it depends on the conditions under which your extracted components can maintain their biological activity. It's best to use a solution that preserves the maximum biological activity of the sample, then dialyze it with a pH 9.6 carbonate buffer. If possible, you can directly use the pH 9.6 carbonate buffer for coating.  2. Does not using a multi-channel pipette affect the results when adding liquid? Shanghai Hengyuan Technical Analysis: For experienced technicians, the impact might be minimal. However, for less skilled individuals, the results could be more variable. That's why many imported ELISA kits recommend using a multi-channel pipette or an autosampler for accurate and consistent sample loading.  3. What is the purity requirement for the antigen? Shanghai Hengyuan Technical Analysis: If the background is not extremely critical, impurities in the antigen may have a limited effect on the coating. In some cases, impurities can even be masked if cross-reactions are avoided (this is only applicable for research purposes). It's advisable to first test different dilutions of the antigen and use an indirect method for detection. 📌 Note: Always dilute your samples before use.
