The summer vacation is already halfway through, and yesterday afternoon, my company's manager took the time to summarize and analyze several ELISA kits that customers frequently ask about. I'm sharing this information here for your reference. 1. When performing tissue ELISA, what kind of liquid is used for tissue grinding? Shanghai Hengyuan Technical Analysis: If you're trying to detect components within a tissue sample, it's recommended to use a buffer with a pH around 7 or lower, and with low ionic strength, to avoid significant interference in the results. If you're using ground tissue for coating, it depends on the conditions under which your extracted components can maintain their biological activity. It's best to use a solution that preserves the highest level of biological activity, and then dialyze it using a pH 9.6 carbonate buffer. If possible, you can directly use the pH 9.6 carbonate buffer for coating.  2. Does not using a multi-channel pipette affect the results when adding liquids? Shanghai Hengyuan Technical Analysis: For experienced technicians, the impact might be minimal. However, for less experienced individuals, the results could be unpredictable. That’s why many imported ELISA kits specify the use of a multi-channel pipette or an autosampler for accurate dispensing.  3. What is the purity of the antigen? Shanghai Hengyuan Technical Analysis: If the background isn't too strict, impurities in the antigen may not significantly affect the coating. In some cases, impurities can even act as a blocker, provided cross-reactions are avoided (this is only applicable for research purposes). It's advisable to first test different dilutions of the antigen for coating and then perform an indirect detection method. ☆ Note: Always dilute your samples before use.
