- Close both the internal and external doors of the sampling chamber and perform a vacuum check to ensure proper sealing.
- Perform the first nitrogen replacement: Insert the rubber tube into the air inlet of the operating room and connect the other end to a plastic bag. Open the nitrogen valve, fill the bags completely, then close the valve and seal the bags tightly.
- Attach the latex gloves to the observation plate flange and secure them firmly.
- Transfer the nitrogen-filled plastic bags into the operating room and allow the nitrogen to fully discharge.
Repeat the nitrogen replacement process for the second time. During this step, the sampling chamber should be evacuated first, and the exhaust valve should be controlled using the foot switch as needed.
For the third replacement, use a mixed gas composed of 85% nitrogen, 10% hydrogen, and 5% carbon dioxide:
- Switch the air path and open the air intake valve. When inflating, first evacuate the sampling chamber using the vacuum pump. Use the foot switch to control the exhaust valve as needed.
- Once the mixed gas is filled in the plastic bag, turn off the air valve and open valve 5. Allow the gas to pass through the flow meter and adjust the flow rate to approximately 10 ml per minute.
- Gradually release the mixed gas from the plastic bag into the operating room.
- After three rounds of gas exchange, the oxygen content in the operating room will be extremely low, creating a suitable anaerobic environment.
Open the deoxidizing agent and activate the deoxygenation catalyst. Turn on the Meilan indicator (Meilan bottle) and observe the color change after one hour to confirm the anaerobic conditions are maintained.
Finally, turn on the UV sterilization lamp to disinfect the interior of the incubator. The sterilization duration can be adjusted based on the specific experiment requirements.
Once the anaerobic conditions are established, proceed with the implantation and cultivation of bacterial strains:
- Check that the sampling chamber door is closed securely.
- Open the external sampling door, place the strain inside, and close the outer door immediately.
- Fill the sampling chamber with nitrogen three times. Start by turning on the vacuum pump and evacuating the chamber to at least 500 mmHg (66 kPa) before each nitrogen flush.
