  5. Perform the first nitrogen gas exchange: insert a rubber tube into the air inlet and connect it to one of the plastic bags. Open the nitrogen valve, fill the bag with nitrogen, close the valve, and seal the bag.
  6. Secure the latex gloves on the observation plate flange and tighten them properly.
  7. Transfer the nitrogen-filled plastic bag into the operating chamber and release the gas completely.
  8. Repeat the nitrogen exchange process for the second time. During this step, the sampling chamber should be evacuated, and the exhaust valve should be operated using the foot switch as needed.
  9. For the third gas exchange, use a mixed gas consisting of 85% nitrogen, 10% hydrogen, and 5% carbon dioxide. Adjust the air path to allow the mixed gas to flow into the plastic bag. Once filled, regulate the flow rate through the flow meter to approximately 10 ml per minute and gradually release the gas into the chamber.
  10. After three rounds of gas exchange, the oxygen level in the chamber should be extremely low. At this point, open the deoxidizing agent and turn on the catalytic deoxygenation system. Monitor the Meilan indicator for color change after one hour to confirm successful anaerobic conditions.
  11. Finally, activate the UV sterilization lamp to disinfect the interior. The duration of sterilization can vary depending on the specific experimental requirements.

Once the anaerobic environment is established, you can proceed with the inoculation and cultivation of microbial strains:

  1. Ensure the sampling chamber door is closed securely.
  2. Open the outer sampling door, place the culture sample inside, and then close the outer door immediately.
  3. Evacuate the sampling chamber using a vacuum pump until the pressure reaches at least 500 mmHg (66 kPa). Repeat this process three times to ensure a stable anaerobic atmosphere.
