How does laboratory instrumentation distinguish different microscopes?

There are several key differences between a biological microscope, a metallographic microscope, and a stereo microscope, particularly in terms of illumination methods, objective lens design, and application scope.

1. Illumination method: A biological microscope typically uses transmitted light, which means the light passes through the sample. It is ideal for observing transparent or semi-transparent specimens such as cells or thin tissue sections. However, it is not suitable for opaque samples. In contrast, a metallographic microscope uses reflected (epi) illumination, where the light comes from the objective lens and reflects off the surface of the sample. This makes it perfect for examining opaque materials like metals or ceramics. Some advanced metallographic microscopes also offer transmitted light capabilities, allowing them to observe both transparent and opaque samples depending on the application.

2. Objective lens design: The high-power objectives of a biological microscope are designed with specific corrections for the thickness of the coverslip (typically 0.17 mm) and the slide (around 1.2 mm). These are often marked with /0.17 for upright microscopes, /1.2 for inverted models, and /- for low-power objectives (10x or below), meaning they don’t require correction. On the other hand, metallographic objectives are usually marked with /0, indicating no correction for glass thickness, as they are used for surface observation rather than through transparent media.

3. Stereo microscopes differ significantly from both biological and metallographic microscopes. One major distinction is their longer working distance—often ranging from 50 mm to 150 mm—compared to the short working distance of a biological microscope, which rarely exceeds 20 mm. This allows stereo microscopes to accommodate taller or thicker objects, such as circuit boards, large components, or mechanical parts, while biological microscopes are limited to flat, thin samples like slides or petri dishes.

4. Another key difference is the depth of field. Stereo microscopes have a much greater depth of field, sometimes up to 10 mm, enabling a larger area to be in focus at once. They also provide a stereoscopic view, giving a three-dimensional perspective. However, their magnification is generally lower, reaching up to around 200x. In contrast, biological microscopes offer higher magnifications, often up to 2000x, but with a much narrower depth of field, requiring precise focusing to achieve clarity.
